Hopefully not! It should be down for a few hours if things go shatteringly well, up to a day if we have to troubleshoot. We tested this upgrade pretty extensively in a remote test bed, so we're feeling confident it should be painless and quick.
Your accounts will come right over, in a basic form. Some things may not transfer over. Paths to avatars, signatures, and private messages will unfortunately not make it. If you have anything stored in private messages that you might want to keep, back it up asap!
